چکیده
Abstract Background The main epigenetic event occurring during the bladder carcinogenesis process is DNA methylation, affecting genes involved in various metabolic pathways and cell regulation. use of biological fluids such as urine sediments could be used a non-invasive approach to enhance cancer management. In this study, we aim determine promoter methylation status panel on tumor biopsies evaluate usefulness samples for assessment. Methods Using methylation-specific PCR technique, explored hTERT , TWIST1 VIM NID2 40 their paired from Moroccan patients. Results tumors showed hypermethylation frequency individual 90%, 85%, 62.5% 72.5% TWIST1, hTERT, genes, respectively. Interestingly, specificity detection was 100% sensitivity detect reached 91.7%; 97.1%; 84% 82.8%, Conclusions Our results clearly show that assessment highly specific has high sensitivity. Furthermore, would useful its potential association with development.

Detection of bladder cancer using novel DNA methylation biomarkers in urine sediments.
BACKGROUND Bladder cancer (BCa) remains a lethal malignancy that can be cured if detected early. DNA hypermethylation is a common epigenetic abnormality in cancer that may serve as a marker of disease activity. METHODS We selected 10 novel candidate genes from the most frequently hypermethylated genes detected by DNA microarray and bisulfite pyrosequencing of bladder cancers and applied them ...
متن کاملA novel set of DNA methylation markers in urine sediments for sensitive/specific detection of bladder cancer.
PURPOSE This study aims to provide a better set of DNA methylation markers in urine sediments for sensitive and specific detection of bladder cancer. EXPERIMENTAL DESIGN Fifty-nine tumor-associated genes were profiled in three bladder cancer cell lines, a small cohort of cancer biopsies and urine sediments by methylation-specific PCR. Twenty-one candidate genes were then profiled in urine sed...
متن کاملDetection of Bladder Cancer Using Proteomic Profiling of Urine Sediments
We used protein expression profiles to develop a classification rule for the detection and prognostic assessment of bladder cancer in voided urine samples. Using the Ciphergen PBS II ProteinChip Reader, we analyzed the protein profiles of 18 pairs of samples of bladder tumor and adjacent urothelium tissue, a training set of 85 voided urine samples (32 controls and 53 bladder cancer), and a blin...
متن کاملCell-Free microRNA-214 From Urine as a Biomarker for Non-Muscle-Invasive Bladder Cancer
PURPOSE MicroRNAs are small noncoding RNAs and microRNA-214 (miR-214) has been associated with the inhibition of cancer cell growth, migration, and invasion. The aim of this study was to investigate whether cell-free miR-214 isolated from urine could be used as a biomarker for non-muscle-invasive bladder cancer (NMIBC). MATERIALS AND METHODS A total of 138 patients with primary NMIBC and 144 ...
متن کاملQuantitation of promoter methylation of multiple genes in urine DNA and bladder cancer detection.
BACKGROUND The noninvasive identification of bladder tumors may improve disease control and prevent disease progression. Aberrant promoter methylation (i.e., hypermethylation) is a major mechanism for silencing tumor suppressor genes and other cancer-associated genes in many human cancers, including bladder cancer. METHODS A quantitative fluorogenic real-time polymerase chain reaction (PCR) a...
